Total RNA Blood Kit (Lab-Aid) is designed for automated isolation and purification of total RNA from fresh whole blood and cultured cells with magnetic particle technology. The product in combination with Lab-Aid 824s/808 Nucleic Acid Extraction System provides high-quality total RNA for downstream applications, such as amplification or other enzymatic reactions. The Lab-Aid 824s/808 Nucleic Acid Extraction systems automate sample preparation, processing up to 24 and 8 samples in a single run, respectively. The product is intended to be used by professional users, such as technicians and physicians who are trained in molecular biological techniques.

Principle

The automated purification of nucleic acids is based on magnetic particle technology, which combines the speed and efficiency of the instrument with the convenient handling of magnetic particles. The purification procedure is designed to ensure safe and reproducible handling of potentially infectious samples, comprising four steps: lysis, binding, washing, and elution.

Lysis: The samples are first lysed with a lysis buffer to release the nucleic acids.

Binding: A binding buffer is added to the lysed samples to adjust the binding conditions. The lysates are thoroughly mixed with magnetic particles to allow optimal adsorption of nucleic acids to the silica surface. The salt and pH conditions ensure that proteins and other contaminants, which can inhibit PCR and other downstream enzymatic reactions, are not bound to the magnetic particles.

Washing: While the nucleic acids remain bound to the magnetic particles, contaminants are efficiently washed away during a series of wash steps.

Elution: Highly pure nucleic acids are eluted in an elution buffer.

This process ensures that the extracted nucleic acids are of high quality and suitable for subsequent molecular biology applications.
