FFPE DNA Extraction Kit (Model: Lab-Aid 824s) is designed for the automated isolation and purification of DNA from formalin-fixed, paraffin-embedded (FFPE) tissues using magnetic particle technology. It is intended for use by professionals, such as technicians and physicians, who are trained in molecular biological techniques. When used in combination with the Lab-Aid 824s Nucleic Acid Extraction System, the kit provides high-quality DNA suitable for downstream applications like amplification or other enzymatic reactions. The Lab-Aid 824s system automates the entire sample preparation process, allowing for the extraction of up to 24 samples in a single run.
Principle
The automated purification of nucleic acids utilizes magnetic particle technology, which combines the speed and efficiency of the instrument with the convenient handling of magnetic particles. This purification procedure is designed to ensure safe and reproducible handling of potentially infectious samples and consists of four steps: lysis, binding, washing, and elution.
Lysis: Initially, the samples are lysed with a lysis buffer to release the nucleic acids.
Binding: A binding buffer is added to the lysed samples to adjust the binding conditions. The lysates are then thoroughly mixed with magnetic particles to facilitate optimal adsorption of nucleic acids onto the silica surface. The salt and pH conditions are set to ensure that proteins and other contaminants, which could inhibit PCR and other downstream enzymatic reactions, do not bind to the magnetic particles.
Washing: During a series of wash steps, contaminants are efficiently washed away while the nucleic acids remain bound to the magnetic particles.
Elution: The highly pure nucleic acids are then eluted in an elution buffer.
